Serialul prezintă povestea intrigantă a lui Michael Scofield, interpretat de seducătorul Wentworth Miller, care înscenează jaful unei bănci pentru a ajunge în aceeaşi închisoare în care se află fratele său mai mare, Lincoln Burrows (Dominic Purcell). Aşteptându-şi rândul pentru scaunul electric după condamnarea în cazul unei crime pe care nu a comis-o, Lincoln se trezeşte coleg de închisoare cu fratele lui, în penitenciarul Fox River. Michael pune în aplicare, pas cu pas, un plan elaborat de evadare pentru el şi fratele sau.
