After the release of Jake Blues from prison, he and brother Elwood go to visit "The Penguin", the last of the nuns who raised them in a boarding school. They learn the Archdiocese will stop supporting the school and will sell the place to the Education Authority. The only way to keep the place open is if the $5000 tax on the property is paid within 11 days. The Blues Brothers want to help, and decide to put their blues band back together and raise the the money by staging a big gig. As they set off on their "mission from God" they seem to make more enemies along the way. Will they manage to come up with the money in time?

După ce sunt eliberați din închisoare, Jake Blues (John Belushi) și fratele lui Elwood (Dan Aykroyd) se duc să viziteze vechea mănăstire care le-a ținut loc de casă de mici. Aici află de la sora Mary Stigmata (Kathleen Freeman) că biserica nu le mai oferea călugărițelor nici un fel de ajutor, iar locul urma să fie vândut și mănăstirea demolată. Singura posibilitate de a împiedica aceste lucruri era ca cei 5000 de dolari cât valora proprietatea să fie plătiți în timp de 11 zile.
Frații se hotărăsc să sară în ajutorul călugărițelor și își reunesc trupa lor de blues pentru a da concerte și a aduna banii pentru taxă. Dar imediat ce pornesc în ceea ce ei cred că este o misiune de la Dumnezeu, membrii trupei își fac mai mulți dușmani decât prieteni și încep să se îndoiască serios de faptul că vor fi în stare să facă o faptă bună.
