Urmându-și visul de a deveni muschetar, D'Artagnan (Logan Lerman) pleacă la Paris, unde îi găsește pe Athos (Matthew Macfadyen), Aramis (Luke Evans) și Porthos (Ray Stevenson) în slujbe mizere, în căutarea unei cauze care să le spele numele. În urmă cu trei ani, după ce au furat un proiect secret dintr-un seif venețian, au fost trădați de partenera lor Milady (Milla Jojovich), care i-a vândut planul ducelui de Buckingham (Orlando Bloom).

În acest timp, diabolicul Richelieu (Christoph Waltz) urzește un plan pentru a-l detrona pe regele Louis (Freddie Fox). Cu ajutorul agentei duble Milady, cardinalul îi înscenează reginei Anne (Juno Temple) o aventură cu Buckingham, sperând că regele Ludovic va crede minciuna. În viziunea lui, lucrurile sunt simple: regina va fi condamnată, Anglia va fi provocată la război, poporul își va dori un conducător mai puternic, iar el va deveni noul rege al Franței.

Pentru a feri Franța de aceste uneltiri, muschetarii se alătură lui D'Artagnan și pornesc în cea mai periculoasă misiune, bazându-se pe spiritul de prietenie care i-a salvat de fiecare dată: toți pentru unul și unul pentru toți!
